Clinical usage | PCR-based methylation tests are used to inform and help predict patient outcomes, as well as serve as a guide for preventive measures, such as surveillance and risk reduction strategies, in individuals with an increased risk of developing cancer. – MLH1: Detection of methylation of the MLH1 gene promoter on FFPE, adjunct to MSI, MMR IHC, and for colon or endometrial tumours demonstrating MSI-H and loss of MLH1 protein expression. – MGMT: Detection of methylation of MGMT gene promoter on FFPE, for prognostic and predictive value for glioblastoma patients. |
Type of sample | – Formalin-fixed paraffin-embedded (FFPE) Tissue/Cell Block (preferred) or – 10 x 5µm unstained slides of FFPE tissue mounted on uncoated slides + 1 H&E stained slide – 10 x 5µm normal tissue slides (If available for MLH1 promoter methylation test) |
Surface Area & Tumour Purity | Minimum surface area of 5 x 5 mm2 per slide. If the tissue area is smaller, please provide additional 5-10 5µm unstained slides. At least 10% tumour purity per slide. |
